Immucilin-H (experimental drug BCX-1777) is a TS analogue that is an inhibitor
of purine nucleoside phosphorylase (PNP).
Changes in the charge distribution from the substrate inosine (bottom left) to the TS (middle) are shown on
the space-filling models (blue - positive; green - neutral, red - negative).
Immucilin-H (right) was designed to mimic the geometry and charge of the TS. It binds to the enzyme 350,000-fold more tightly than inosine. (Miles et al. (1998) Biochemistry 37, 8615-8621).
